After making a change in a sequence file and saving that file (it still has the same name but a new saved date), do I .......

Need to add that 'revised' sequence to the Show file and delete the old sequence?  Or does the CP automatically know that the 'revised' sequence is still the one to use in the Show?

As long as the name is the same, you don't need to do anything.  The show player will see the new revision and process accordingly.
